A new clinically-relevant rat model of letrozole-induced chronic nociceptive disorders

چکیده

Among postmenopausal women with estrogen receptor-positive breast cancer, more than 80% receive hormone therapy including aromatase inhibitors (AIs). Half of them develop chronic arthralgia - characterized by symmetric articular pain, carpal tunnel syndrome, morning stiffness, myalgia and a decrease in grip strength which is associated treatment discontinuation. Only few animal studies have linked AI to nociception, none arthralgia. Thus, we developed new AI-induced nociceptive disorder model mimicking clinical symptoms induced AIs, using subcutaneous letrozole pellets ovariectomized (OVX) rats. Following plasma dosage at the end experiment (day 73), only rats least 90 ng/ml were considered significantly exposed (OVX + high LTZ group), whereas treated animals less pooled OVX low group. Chronic set rapidly was maintained for 70 days Furthermore, saw no alteration locomotion, or experimental anxiety during this period. Bone parameters femora altered all compared Sham+vehicle pellet. A mechanistic analysis focused on TRPA1, receptor suspected mediate AI-evoked showed modification its expression DRG. This long-lasting rat model, efficiently reproduces affecting patients' daily activities quality-of-life. It should help study pathophysiology promote development therapeutic strategies.

A Clinically Relevant Unilateral Rat Model of Parkinsonian Akinesia

In transplantation research on Parkinson's disease, behavioral evidence of graft-specific improved function is as necessary as anatomical or chemical data in evaluating the success of the procedure. The circling model of Ungerstedt has been widely adopted to provide such evidence, in part because it is so convenient and (relative to primate models) inexpensive. In this model 6-hydroxydopamine (...

Differential Effectiveness of Clinically-Relevant Analgesics in a Rat Model of Chemotherapy-Induced Mucositis

Chemotherapy-induced intestinal mucositis is characterized by pain and a pro-inflammatory tissue response. Rat models are frequently used in mucositis disease investigations yet little is known about the presence of pain in these animals, the ability of analgesics to ameliorate the condition, or the effect that analgesic administration may have on study outcomes. This study investigated differe...

Chronic heterogeneous sequential stress increases formalin-induced nociceptive

Abstract Introduction: Chronic heterogeneous stress may be better for evaluation of the effect of chronic stress situations on the nociceptive behaviour. The present study investigated the effects of chronic heterogeneous sequential stress on thermal-induced nociception and formalin induced pain behavior in rats. Methods: In the present study, adult rats (220-300 g) were used. Animals were ...
